ergänzende Hinweise | Throughout the years DNA Origami was both used as a tool for fundamental physical measurements and heavily researched for a wide variety of biomedical applications. Here we offer an insight in a cross-section of the two by determining different binding parameters, such as the Gibbs free energy, of a antibody-functionalized DNA Origami bound to the mammalian cell surface. Within this lab course the students will learn how to produce and purify state-of-the-art fluorescently labeled DNA-Origami functionalized with cell surface antigen- targeting proteins, alongside the quality control methods like agarose gel electrophoresis and chromatography. The students will also be given an insight into the aseptic techniques necessary for work with the mammalian cell culture and the preparation of the cells for the assays. Subsequently, produced DNA Origami structures will be used in a cellular binding assay and analyzed in a flow cytometer. Finally, the concepts of data analysis will be presented in order to determine the binding energy and other physical parameters of interest. |
